A preorganized β-amino acid bearing a guanidinium side chain and its use in cell-penetrating peptides.

چکیده
A cyclic β-amino acid (APC(Gu)) bearing a side-chain guanidinium group has been developed. The APC(Gu) residue was incorporated into an α/β-peptide based on the Tat(47-57) fragment, leading to an oligomer with substantial helicity in methanol that enters HeLa cells much more readily than does the corresponding Tat α-peptide.

Evaluation of the Effect of Less Negatively Charged Amino Acid Substitution in Synthetic Tetramer Peptide S3 Derived from Horseshoe Crab Ambocyte on its Antibacterial Properties
Introduction: The study of the effects of synthetic peptides with antibacterial properties can provide more effective antibiotics. This study designed, expressed, and investigated the Sushi 3 tetramer peptide. Subsequently, it was compared in terms of changing antibacterial properties with another Sushi3 tetramer peptide the aspartic acid and proline amino acids of which were replaced with glyc...
متن کاملHeLa cell entry by guanidinium-rich beta-peptides: importance of specific cation-cell surface interactions.
Short cationic oligomers, including arginine-rich peptides and analogous beta-amino acid oligomers ("beta-peptides"), can enter the cytoplasm and nucleus of a living cell from the extracellular medium. It seems increasingly clear that multiple entry pathways are possible, depending upon the structure of the guanidinium-rich molecule, the type of cell, and other factors. We have previously shown...
متن کاملCell penetrating and transytosing peptides: powerful strategies for oral insulin delivery
Insulin is essential for type 1 and advanced type 2 diabetes to maintain blood glucose levels and increase the patient’s longevity. Frequent subcutaneous insulin injections are usually associated with pain, local tissue necrosis, infection and nerve damage. Recently, a number of new delivery methods such as oral insulin delivery have been developed to overcome these limitations and increase pa...
متن کاملNew Charge-Bearing Amino Acid Residues That Promote β-Sheet Secondary Structure
Proteinogenic amino acid residues that promote β-sheet secondary structure are hydrophobic (e.g., Ile or Val) or only moderately polar (e.g., Thr). The design of peptides intended to display β-sheet secondary structure in water typically requires one set of residues to ensure conformational stability and an orthogonal set, with charged side chains, to ensure aqueous solubility and discourage se...
متن کاملSynthesis of C-linked carbo-β2-amino acids and β2-peptides: design of new motifs for left-handed 12/10- and 10/12-mixed helices.
C-linked carbo-β(2)-amino acids (β(2)-Caa), a new class of β-amino acid with a carbohydrate side chain having d-xylo configuration, were prepared from d-glucose. The main idea behind the design of the new β-amino acids was to move the steric strain of the bulky carbohydrate side chain from the Cβ- to the Cα-carbon atom and to explore its influence on the folding propensities in peptides with al...
